| PioPlus is combination of 3 drugs for the management of Type II Diabetes Mellitus when dual therapy is not sufficient to achieve the desired sugar levels. |
Composition:
Glimeperide 1/2mg + Metformin Hydrochloride 500mg (SR) + Pioglitazone 15mg |
| Mechanism of action |
Glimepeiride stimulates the insulin release from functioning pancreatic β cells and inhibits gluconeogenesis at hepatic cells. It also increases insulin sensitivity at peripheral target sites.
Pioglitazone is a thiazolidinedione oral antidiabetic agent, which acts primarily by decreasing insulin resistance. Metformin Hydrochloride is a biguanide oral anti-diabetic agent, which mainly acts by decreasing hepatic glucose output and increasing glucose utilization in the liver & adipose tissue. |

| Side effects |
Pioglitazone : Headache, URTI, Myalgia, Sinusitis, Pharyngitis, Anaemia, Oedema.
Glimepiride: Vomiting, G.I Pain, Diarrhea, Pruritis, Erythema, Urticaria, Morbilliform, Leucopenia, Agranulocytosis, Thrombocytopaenia, Haemolytic anemia and pancytopenia, hyponatremia, Blurred vision, Jaundice.
Metformin: Abdominal distention, Nausea, Vomiting, Diarrhea, Flatus, Oesophageal obstruction. |
| Precautions |
Heart failure may develop or be exacerbated due to fluid retention
Edema; may initiate or worsen edema
Concurrent use with alcohol; risk of hypoglycemic episodes
Liver enzyme elevations, mild; risk of exacerbation

| Contraindications |
Heart failure, New York Heart Association Class III and IV
Hypersensitivity to pioglitazone, glimepiride, metformin
Diabetic ketoacidosis.
